Extraction of Caffeine from coffee by Homogenous Liquid-Liquid Extraction (HLLE) method and determination with Spectrophotometric UV-Vis

Caffeine is the most popular stimulant in the world that stimulates the central nervous system of the human body and is added manually in some products, including energy drinks. Caffeine content is very variable in food. Caffeine is a xanthine crystalline alkaloid that naturally exists in tea, coffee, cola and in small quantities in cocoa. The extraction of caffeine in coffee has always been a concern for researchers and industry owners. The homogeneous homogeneous liquid-liquid micro-extraction of the homogeneous solution containing the sample and the extractor phase is converted to two-phase solution by changing the conditions governing the solution and the extraction phase is deposited. In recent years, the use of mathematical models in the chemical and food industry has been considered to improve performance and increase efficiency. The Response surface Method (RSM), with the lowest data, can determine the optimal value of several variables simultaneously [1]. In this study, the combination of liquid-liquid-micro-extraction method (HLLE) with ultra-violet spectroscopy was used to extract caffeine in coffee and using the software, the response of the obtained data was analyzed. Variable parameters after optimization were extract extraction pH (3.9), extraction temperature (40 °C) and concentration of extraction phase of triton x-114 (1%). The calibration curve of the method was drawn. The relative standard deviation of 5.0%, the coefficient of correlation of 0.996 and the linear range of method 0.1-2 μg.mL-1 were determined. This method has been used to measure caffeine in a sample of coffee bean with two ways coffee was smoked and not smoked.
